Shan Masood was unbeaten on 88 off 137 balls, including 11 fours, anchoring the innings alongside Imam-ul-Haq, who made 63 as the pair added 155 runs together.
Pakistan’s reply got off to a poor start when opener Azan Awais was bowled by Kemar Roach for a nine-ball duck.
Salman Ali Agha was unbeaten on nought off 9 balls at the other end when play ended for the day.
West Indies had been dismissed for 311 in their first innings, with Mohammad Ali taking four wickets for Pakistan.
Pakistan will resume their first innings on day three, still trailing by 112 runs.
